RAA · Recombinase-Aided Amplification

Isothermal nucleic acid detection. Results in 5–15 minutes.

RAA runs at a constant 37–42 ℃ with no thermal cycler. Lyophilised reagents and a portable fluorescence analyzer bring molecular-level testing to the bench, the plant floor and the field.

01 · Technology

RAA replaces thermal cycling with enzymes.

Recombinase-Aided Amplification is the isothermal amplification platform behind every ZC Bioscience reagent. All kits are developed and manufactured in-house.

How it works

A recombinase obtained from bacteria or fungi binds tightly to the primer DNA at ambient temperature, forming a recombinase–primer complex. When the primer locates its fully complementary sequence on the template, single-stranded DNA-binding protein helps open the double strand, and DNA polymerase synthesises a new complementary strand. The product accumulates exponentially.

With a labelled fluorescent probe the reaction can be read in real time; a fluorescence result is typically available within 5–15 minutes. Reagents are pre-mixed and lyophilised in the tube: add Buffer A, Buffer B and the extracted sample nucleic acid, then read on the Genchek analyzer or a standard real-time instrument.

STEP 01

Recombinase binds primer

Recombinase–primer complex forms at ambient temperature.

STEP 02

Target search & strand opening

Complex scans the template; SSB stabilises the opened strands.

STEP 03

Polymerase extension

Strand-displacing DNA polymerase extends from the primer 3' end.

STEP 04

Exponential readout

Probe cleavage gives a real-time fluorescence curve in 5–15 min.

Simple operation

Molecular-level testing that does not require specialist staff. A two-step workflow: extract, then amplify and read.

Fast

Results in 5–15 minutes with a fluorescent probe; gel-readable product within 30 minutes for basic kits.

Accurate

High sensitivity and specificity — the recombinase–primer complex recognises the target sequence by base pairing.

Portable

Compact instruments suited to harsh field conditions and on-site inspection — the Genchek analyzer weighs 3.8 kg.

Isothermal

Runs at 30–42 ℃ (optimum 37–39 ℃). No thermal cycler or other expensive equipment required.

Lyophilised

Reagents supplied as freeze-dried pellets: ready to use, easy to transport, and free from freeze–thaw loss.
